- Half-life in the body: Circulating 25-hydroxyvitamin D [25(OH)D], the main storage form measured in blood tests, has a half-life of approximately 2–3 weeks (typically 15–25 days). - Best time of day to take it: With a meal that contains dietary fat (e.g., breakfast, lunch, or dinner) to maximise absorption. Morning is optimum.
- Optimum dose: Highly individual. Official RDAs are 600–800 IU/day for most adults, with a tolerable upper intake level of 4,000 IU/day for long-term use without monitoring. Many functional and research-oriented sources suggest 2,000–5,000 IU/day (or higher under blood-test guidance) to achieve and maintain serum 25(OH)D levels in the 40–60 ng/mL (100–150 nmol/L) range for many adults. - Long-term safety: Vitamin D3 is generally recognised as safe at recommended doses. Toxicity is rare and almost always linked to prolonged very high intakes for months, leading to hypercalcaemia. Doses up to 4,000–10,000 IU/day have been used safely in long-term studies when monitored.
